Salicylic acid perturbs small RNA-gibberellin regulatory network in immune response of potato to Potato virus Y infection and renders plants tolerant to the pathogen

چکیده
18 Potato virus Y is the most economically important potato viral pathogen. We aimed at 19 unraveling the roles of small RNAs (sRNAs) in the complex immune signaling network 20 controlling the establishment of tolerant response of potato cv. Désirée to the virus. We 21 constructed a sRNA regulatory network connecting sRNAs and their targets to link sRNA 22 level responses to physiological processes. We discovered an interesting novel sRNAs23 gibberellin regulatory circuit being activated as early as 3 days post inoculation, before viral 24 multiplication can be detected. Increased levels of miR167 and phasiRNA931 were reflected 25 in decreased levels of transcripts involved in gibberellin biosynthesis. Moreover, decreased 26 concentration of gibberellin confirmed this regulation. The functional relation between lower 27 activity of gibberellin signaling and reduced disease severity was previously confirmed in 28 Arabidopsis-virus interaction using knockout mutants. We additionally showed that this 29 regulation is salicylic acid dependent as the response of sRNA network was attenuated in 30 salicylic acid-depleted transgenic counterpart NahG-Désirée expressing severe disease 31 not peer-reviewed) is the author/funder. متن کاملInduction of Resistance to Potato Virus Y (PVY) Using Hairpin Construct of Coat Protein
Potato virus Y (PVY) is one of the most damaging viruses of potato plants which infecting most cultivars and causing significant yield and economical losses. The application of the concept of pathogen derived resistance opened new horizons for the development of virus-resistant plants. This research was carried out to study RNA silencing to engineered potato plants that are resistant to potato ...
متن کاملInvolvement of Potato (Solanum tuberosum L.) MKK6 in Response to Potato virus Y
Mitogen-activated protein kinase (MAPK) cascades have crucial roles in the regulation of plant development and in plant responses to stress. Plant recognition of pathogen-associated molecular patterns or pathogen-derived effector proteins has been shown to trigger activation of several MAPKs. This then controls defence responses, including synthesis and/or signalling of defence hormones and act...
متن کاملDynamics of Responses in Compatible Potato - Potato virus Y Interaction Are Modulated by Salicylic Acid
To investigate the dynamics of the potato-Potato virus Y (PVY) compatible interaction in relation to salicylic acid-controlled pathways we performed experiments using non-transgenic potato cv. Désirée, transgenic NahG-Désirée, cv. Igor and PVY(NTN), the most aggressive strain of PVY. The importance of salicylic acid in viral multiplication and symptom development was confirmed by pronounced sym...

One of the most efficient mechanisms by which plants protect themselves from invading virusesis the specific RNA-dependent silencing pathway termed post-transcriptional gene silencing(PTGS). In this mechanism, resistance to a virus is engineered through the expression of asegment of the virus genomein transgenic plants. Potato VirusY (PVY) is one of the mostdamaging viruses of potato, infecting...
